@unk0damnation For me, it's not so much that she's murderating people atm, it's that we haven't been given sufficient excuse for her to go from zero to 11 instantly on the psychopath scale. She knows she used to be human so she's shown an aversion to harming people. Hopefully this is explained later as mostly the effect of Taboo, etc.
@Aereus Well, there's no 'zero' that we've known. We only 'know' she used to be human, but what kind of human? She only expressed her concern about 'maybe I should communicate with human, but with this body, how?' a couple times, which totally failed in chapter 1 or something close when her home was burned by random adventurer. That's it - what was she like as a human? From the dialogues, she sounds like a game nerd - think stereotypical JP nerd: she's likely antisocial? Maybe? I don't know, you don't know. Why assume she has 'aversion to harming people in the first place' or whatever moral standard she would have in the first place?
It was around this point in the web novel that I started to lose interest in the story.
Like @Aereus felt, I too thought Kumo-ko's sudden willingness and complete lack of reservation in killing humans came out of nowhere, and it revealed a major problem with the story:
We don't know anything about kumo-ko. Her history, or her personality. Everything before this point was dungeon grinding and skill unlocking and that was fun and enjoyable. It also covered up the lack of any real detail on Kumo-ko's character. But now that flaw gets forced into the spotlight and once it's revealed, you can't really ignore it. If we had some character development or at least some hint of her personality, then her actions would make sense.
If you thought Kumo-ko's character made no sense or felt too forced now, be warned it's going to get A LOT WORSE from here on out. And after a certain point, I quit the web novel out of sheer frustration at how Kumo-ko's actions made no sense in regards to her character.
Maybe the manga might go in a different direction, but so far they've followed the original story pretty closely, and that's not a good sign.

Since that chapter we know that she hates the people that burns her homes, also the full course of poison is certain dead so she always had the intention to kill them.
She clearly wasn't joking on both momments, the real problem is that the manga artstyle makes funny almost everything she says so it didn't felt like a threat.

By this point we know a lot about Kumoko personality.
She's a cheerful girl who like to eat food and sweet things. She hates to work hard for anything, unless she doesn't have any other option. She has a really big sense of Pride over herself and her work. She would like to live her life the most relaxed as posible but her current enviroment doesn't let her do it. She hates everything about interacting with other people and will evade it as much as posible, and doing pointless things.
Kumoko usually tries hard to see the good side to every situation and tends to become overconfident with time, only to end extremely frustrated when something doesn't go the way she expected.
Kumoko only wanted to comunicate with humans because it was the best way to obtain good food and sweets.
Later is revealed that she doesn't have empathy. Kumoko only thinks about herself or a little group of persons she truly trust. And she's truly grateful towards those who have save her.
After the events of this chapter she stops seing herself as human anymore and fully accepts that she's now a monster.
How does being a monster and kill some humans affect her personality when we already know that she doesnt feel guilt? And even before obtainig the title she already showed amoral behavior towards monster.

Did you skipped the part of the novel where it's explained that the soul tends to adapt to it's new body? You can clearly see how Kanata unconsciously starts to behave and think like a woman. The same can be applied to Kumoko and even Heresy Immunity can't protect her from this because this change is "Soul Behavior" not an attack from the System

Also Did you reach the part where it's revealed that Kumo wasn't a human on Earth, but a normal spider that lived inside the classroom? And the real Wakaba Hiro is D.
Kumo behavio can be also explained as spider behavior
* Hikkikomori = A house spider never leave her web
* No parent = Spider usually get separate from their parents right after they birth
* Doesn't want to relate to others = Spider are lonely creatures
* Lack of empathy = Spiders are natural hunters/murders
* And many more
This is why she doesn't have/remember her past life, she only have the school memories that D gave to Kumo to act as her decoy. D always expected Kumoko to die almost
