Isaac is far too smart, he already figured out that Skelly is stuck in a timeloop just from a few words. Even so he couldn't be absolutely sure of it, so him betting his life on it is pretty crazy.

There is a good chance that Mulus isn't even human, just look at his pupils. Him being a very pretty man isn't so strange. He is also tall and has broad shoulders, so unless the artist is just messing with us it doesn't make sense for him to be a woman.
