And it was cheap effort for the author for trying to make it look like that b*stard knowing about MC's demon eyes seem like something dangerous for MC, even though that b*stard himself said in previous chapter that humans are also able to use demon eyes.

He seemed to possess demon eyes himself. That being said, what are the conditions for humans to be able to use them? Possibly it's something that could be considered suspicious, so while it might not alone mean anything, if you pile it on top of other evidence of foul play, it could mean something. The hero's underling in the previous chapter did explain how people around Elixir keep dying once Elixir doesn't have any further use for them. Just Elixir messing with the hero's own plans concerning Idly (his vessel) should be enough for the hero to come and check out Elixir. Trying to suddenly pressure Elixir about the demon eyes would be a standard interrogation technique to drive Elixir into making a mistake.

Cross-shaped pupils in the very last image of the previous chapter. What else could it mean? Together with his talk about a vessel, I theorised maybe his particular demon eyes are connected to being able to transfer his soul into a younger body, granting an immortality, after a fashion.

His parents literally have parts of the demon queen sealed inside their bodies, it's not that much of a stretch to think that the demon eyes could have come from that. He already said that humans can have demon eyes even if it is rare.
