Dont think so. From Doylist point the mangaka had not yet come up with Butterfly and did not have him in mind when writing this story... From Watsonian well, to avoid spoilers for the final chapters, I will just say that the Major, judging by everything, was a regular military man or maybe a mercenary (this was not specified), assigned to perform a specific task (to exterminate Desert Rabbits) and his ego was so hurt by Jesus' feat that he took personal revenge. Butterfly intervened afterward extermination operation. After all, most of the Butterfly's followers are marked with the sign we know, and Major doesn't have it
