Where did the pre-schooler sisters get enough insight into relationships and life that they'd try to protect their older sister doing this sort of spiel, and then also the suspicion that she's actually a closeted homosexual..?

Despite all their prodigy-like behavior they still approach it in such a tactless way considering the sensitive nature of both the closeted-ness and the current status of said relationship; are they trying to sabotage, or could it be that they know it will end up in a positive outcome in the long run despite potentially causing their sister to go deeper into the closet and/or damaging the relationship between everyone involved in the short term?!

So many questions regarding these magical little girls that need answers! Have they 5D-chessed the future, are they just savants in certain areas but dumb in others, could they be maverick loose canons that don't care??!?!!!

Oooor maybe it's just a tired standard trope that vaaaaguely drives the romance progression

Imo you have played the 5D-Chess against yourself and have interpreted way to much into their behaviour and their understanding about relationships.
They know only the close family way of love and what you do with your sisters, holding hands, hugs and cheek kisses, nothing to special but shows closeness.
Now that their sister who never has shown these closeness or even a bit of friendship to anyone except them, has brought someone home over the weekend, so they are just a bit suspicious and probably a bit annoyed and want to know if Mashiro is "worthy" of taking their sister away to spend the weekends instead of them. That is not necessarily romantically, only if you want to see it with pink coloured goggles, then you can interpret it into it and your complete point would be true but for me they are just two innocent kids who don't know a thing about romance and even more homosexuality, just like everyone else at their age, only with a hint of sisconnes, as they were probably spoiled by her their whole life.
But in any way I agree it kinda is a trope, that someone bursts in and forces progress (thought that already with the implied relationship between nurse and prez, but we never got a reaction to that so far, besides the initial wtf). Nagi will think about if she wants to kiss Mashiro, just like she already figured that hugging feels different from person to person.

Sorry, I know it was a long time ago since you have written that and my answer is long and in shit english but I wanted to make clear that we didn't need to see romantical behaviour everywhere, especially from very young kids, they don't know a thing about it and just would get very confused by all of it.
