His expression on page 22 reads to me as someone fond of a child, like a sibling. The one on page 26 I'm less certain about. It's possible that it's a serious face, concerned for this child's safety? since young Towa seems to have less boundaries or the wariness to be more careful around Griz. We can presumably trust Griz around young children but what about other people?
I highly doubt that Griz had a hand on the holy mark's appearance. My personal theory is that it wasn't time for him to get the mark until he was older and came back for the second time

Griz is scaring me (mainly cause if the implications of their old age gap), i dont know how to feel about him. The way hes extremely affectionate is strange. Even if he regretted not protecting the kid, that wouldnt explain it
