No. The plan was to wait until both sides were in the middle of their DragonballZ style kamehameha-fest in chapter 1, take advantage of their preoccupation and kill both of them in one go. The problem was that Naga and Anastasia's ultimate magic powerscaling battle caused that big explosion that isekai-ed all of them and caught the twins by surprise as well.

I find their backstory a little hypocritical; so they obviously dislike humans for mistreating them towards the end of their time with humans, but humans dislike demons for the wars and being slaughtered by the demon king. It's just circular logic and hate. Then again, it hasn't actually said what any side is doing with the humans, all of them may be ignoring the humans, the demons included - and perhaps that was intentional on their end, given their mother.
