The demons are at way too much of a disadvantage here. Given how effortlessly she defeated the doggo in chapter 1, she's at least a relatively strong demon, but...

The prayers of a first day saintess-in-training are enough to make her collapse, a simple medical herb or potion causes significant injury, and they don't seem to have healing magic of their own.

They need to show something that the demons have an advantage on, otherwise this war is going to end in a completely one-sided loss.

For the spell, I think it's specifically casting the spell that did so much damage, since she was both the epicenter and the conduit. Like holding a flashbang in your mouth when it goes off.

For the lack of healing magic, I think it's specifically human healng magic they can't use. A lot of the time settings like these have demons healing on contact with miasma or chaotic energy or darkness or some other such excuse element, but suffer injury on contact with holy/healing magic.
And we see our demonsaintess heal with blood here. So demons have that edge – their opponents are walking bottles of healing potion.

...as Akuno stated, she can use "Blood of the Innocent" - so even if that's a thing, there's no guarantee that your average adult human qualifies in terms of being innocent.

Regarding the matter of taking damage from holy power, like with those herbs. I also wrote in the last chapter comments that I wonder why the holy relics on her wrists don't burn her to ashes, but let's assume those bangles are designed in a way that they will not harm whoever wears them. Still weird, considering that any other contact with holy power seems to deal heavy damage to Akuno but let's leave it at that.

Now, why would she not simply wear gloves when handling the herbs? That would be because creating a healing potion is a magical procedure. No way you simply squeeze herbs and get a potion from thin air. It's not explained but by what is shown in the panels it's indicated that inducing your magic power into the herb is an essential step in the process. I would imagine that doing this is a delicate procedure and if you just fire your Mana remotely without proper guidance, you might as well just turn the herbs to cinders. Or she needs to channel holy power received from the divine through prayer into the herbs to amplify their healing effects. Who knows. In both cases it would make sense that this energy transfer requires direct physical contact.
