I was wondering this too, so I checked the raw and it turns out this was wordplay hence why he didn't catch it. In JP she said "Kouhai oneechan to seishun shimasho!"
The thing here is "seishun" (青春) means youth (literally "green spring") So he likely heard her say "Experience youth with your kouhai oneechan!" But she swapped the 青 with a different sei (性) which is the kanji used for sex.
It's a joke that doesn't work in English.
