Until now, the Bo/Kaoru plotline was that Kaoru had a bad reputation as a shallow flirt(which she was) and Bo shrugged her off as insincere. As of this chapter, Bo was actually into her the whole time and just pretending like she wasn't, and that is a tsundere retcon. (Or kuudere, technically).

Chapters 174 188 and 189 are particularly important for what you're talking about.
It isn't just about Kaoru though. Kaoru had her growth moment. Mask/Bou/Mayo still needed hers. In the beginning she was denying even the possibility of romance believing the only way she could draw manga was to pour everything into it. She wasn't refusing Kaoru's advances just because she thought of her as frivolous. Mayoi doesn't say she has been in love the whole time, just attracted. Like, she uses that exact word (pg 9).
