Oh, this is a great question!

It's a verbal tic that a mushroom speaking Japanese might have. Essentially it's kind of a catchphrase.

(キ)ノコ = "Mushroom"

As an illustrative example: take Hololive's Pekora, whose character ends sentences with ぺこ(Peko).
Or how instead of saying こんにちは (Konnichiwa) she says こんぺこ (Konpeko)
