Method 4 doesn't work. At least based on the last chapter, if the Mandragora dies while still buried, the poison won't be neutralized and it'll spoil.
To remove the poison it has to be dragged into the sunlight while alive, so you can't kill it while it's buried. As for the other method, I imagine you could set up like a snare trap, if you don't have a long enough rope, and just catapult the mushroom out of the dirt by setting it off, but I wonder if it can still scream a second time if you don't finish it quickly....
If exposure to the sun removes all the toxins/poison from the body, then in theory it shouldn't matter if you kill it above or below ground according to the book's explanation.
