It exists but its not common available in mass how is today, characters even pointed out what he is doing meaning they know, its just not common enough to have a resistance of sorts due to lack of exposure, after all mocking important people in public can lead to a swift death, similar like today comedians can get canceled if they talk about certain topics and certain type of people when people are exposed to that, they find it even more funny due to lack of continued exposure.

I am sure they exist, the author is not even making such claim, just that in this scale and in this situation its going to have a bigger effect because its unheard of, after all he is doing this in the Coliseum during a fight, this is why "Marvel humor" originally worked because people werent expecting comedy during what was supposed to be a serious situation and also why it later failed because of continued exposure were it became the norm.
I would say that pre-modern humor was more based on something simple being unexpected or different from norm. Look at clowns who are now seen as scary, but used to be funny because they had weird makeup and too big pants. Or medieval custom of majing a day where jester pretented to be king. People found humor in simple or merely weird things, and didn't care if it was something they've seen before or not sophisticated enough. If someone gives a performance, people would laugh because hey, something funny happens and that's enough, life is boring and shitty as it is, so every occasion to laugh is a good one.

With this, I can at least that he had better chance with his comedy with medieval-like audience, since they would find funny even just the fact that guy starts making jokes in middle of fight, and wouldn't say stuff like "Meh, unfunny boring and unoriginal. Totally mid, only NPCs would laugh at that" like modern people could. Basically, people weren't spoiled by memes, cynism and "ironical" humor, giving him better chance.

Are you being serious? Comedy has existed in society for as long as society itself. We’ve seen graffiti in Pompeii ruins that attests to that.
You cant possibly compare that shit to what weve developed comedy into. We are objectively funnier than everyone before us you can stare at your pompeii scribble and giggle to yourself ill be watching big chungus. Tuh

When I was in highschool I know someone who thought TV was invented before 19th century. And I have seen adult westerner that think people outside Europe was uncivilized random tribes before colonization
Except that he was kinda right. People just misinterpret his words. He said that comedy is rare not that it doesn't exist. If you think about the setting. It make perfect sense considered theatre were rare back then and the primary entertainment was gladiator's fight

you misunderstand what that means, mostly because of a misunderstanding on the translator's side.

by not being developed he means that it's not as common in that world. in the modern era, everyone watches comedy basically every day in some form. in medieval periods (and before) comedy was mostly something that happened every month at most, and street performers weekly at most. also information back then was not that easy to access, which makes it hard to make jokes that everyone understands. it's not like the mangaka is saying that in old periods comedy wasn't developed at all, but it wasn't as developed as in modern times, obviously. it's just that the way the translator did not translate it correctly, and 'Things like comedy aren’t developed in this world' is not the right way to translate what the mangaka wrote. it's the translator's fault.

Fantasy and time travel manga seems particularly prone to this sort of thing. "These primitives haven't even developed refine humor, they don't know about showing the same dumb facial expression five times in a row after someone else pretends to trip! They don't even have goofy sound effects!"
If you look at comedy as a science, it's actually true though. Yeah, every culture throughout history has had knee slappers but the purpose of professional comedy and comedy routines is to insure that you have a consistently positive reaction and can string together a series of jokes, skits or gags for maximum effect.
