Well the mage association was the one who set up the anti-skill barrier, and are in charge of bestowing power onto the heroes. So if glasses wasn't willing to do it they likely have a tool handy that can do something similar. Especially since they seem shady enough to be a later plot villain.

Yes, its just her, but knowing that they can just make it a daily thing. If she refuses it would mean he got her. And her casting it would be hard to fake as it has visual effects. So its both a way to check and lift it. So yes its only her, but that doesn't really matter.
All in all his ability is wildly powerful and useful. He can control any female of any species. Say he goes and touches a sleeping dragon. Its a crazy powerful taming ability. Yet they just kneejerk reaction with a you must die for having that power.

He could have gotten her under his skill after that night with her, sayaka herself says that she'd completely succumb to his skill if she didn't actively resist and cast dispel before the effects took hold deep in her mind.
Kirishima decided not to use on her since she already loved him deeply enough, and felt it wasn't really needed. Probably felt bad for her.
