Counterpoint: if she's ACTUALLY immune, she might be a new kind of character--one who can step back and observe the situation and make sparks fly for fun. Imagine if she becomes Sou's friend and starts feeding him misinformation to make his misunderstandings WORSE.
Ruri is as immune to Soy Boy as Ryuen is immune to Ayanokoji's charms.

It's hard to say, honestly. I do think the class A guards slightly edge it out for me maybe purely because I love their designs so much. Dark hair and beauty mark combo of the Konno sisters is my favorite anime girl design alongside ponytails while Kakizaki is just so cute with her lowkey slyness. But I really like Kanaria and Hinata's "gochisou-sama deshita" moment really raised her stock up. Miku has kinda become too flanderized though.
R18 Sou is also a worse person than normal Sou. The girls frequently point out how he talks to women like equals and how he's kind to them unlike literally every other man on earth, whereas r18 sou doesn't really show that.

I mean I just remember when he was taking miku to bed, it's her first time and he basically tells her he thought she was a slut because she looks like one. He kind of seems like a dick.

Regular sou on the other hand is basically a friendly puppy dog.
I would've preferred if the R18 version was basically the lewd "what-if" fantasies the girls were having.

And everyone wondering or wanting the new girl to be immune to Sou are reading the wrong series. C'mon now, we're too far into the series to not know what it's all about.

Let's define an "Otokonoko" roughly as a boy who dresses and acts in a way that he's easily mistaken for a girl. What would that imply in the 1:39 world? One possible option is what I mentioned. A guy who does it for self-protection, so that girls around him wouldn't jump on him when he's alone with them. Hard to pull off if girls could sniff him out by scent, but maybe possible.

Then again, girls in that world are the aggressively horny ones - given how guys react to them, an Otokonoko in that world would likely be gay, maybe aggressively so. So being an Otokonoko could mean someone who's lusting after males like a horny girl, and thus Otokonoko might be considered "just as dangerous" from a boy's perspective.

But within a so-called Flower Garden (where males are among themselves) an Otokonoko who dresses and acts like a girl would probably be quite the disturbing factor. In that case it could be a bully who picks on other guys by imitating a girl's behavior thus instilling a similar kind of fear.

This world has options to have such a character, but a normal Otokonoko wouldn't fit in.

But I highly doubt we will see anything like that anytime soon.
