Will the Count be the next big bad in this arc? It's a tall task to make a character who's not inherently evil to be a villain, but I suppose his lack of empathy towards others to reach his goals is a good starting point.

The Aira, Jiji stuff will climax here since Okarun and Momo are about to get together. They had to settle those storylines eventually. You should be happy that they're doing both at once since that seems to suggest it's an intentional push for a conclusive ending to the unrequited love stuff soon.

The Nurse "Queen" first and last appeared in Chapter 25.

As for Count, he first appeared as a flashback in Chapter 120, and then in the next Chapter he appeared himself.

It's funny how Queen got the ability to see spirits by just... looking at exorcisms? It does make sense considering exorcisms are 100% the real deal in their world, so I suppose visiting enough of those will expose you to sufficient spiritual energy.
