It's been some time since those chapters came out, I guess. People may have forgotten that both Gotou and the kouhai here have done and/or said some pretty questionable stuff earlier. No, he doesn't know that Mishima loves him. Despite all her obvious hints and her even ultimately saying she likes him, he doesn't understand she meant that romantically. This is one of those frustrating quirks about this (and many other) main character(s). You need to be extremely upfront with them, and even then it's a coin toss whether they get it or not.Yoshida is callous enough to (unintentionally or otherwise) show so much affection for Sayu in front of Mishima knowing full well how she feels about him.
Also, at the start she was kinda showed as a Kouhai who makes mistakes in her work to spend more time with him (with how she makes ton of mistakes, but suddenly when there's a deadline she doesn't make any, plus the fact that she said that she don't want anyone else than him to teach her)No, he doesn't know that Mishima loves him. Mishima didn't clarify her feelings are romantic, thus Yoshida isn't remotely aware of her feelings for him. It's the usual tropey song and dance where the girl does almost everything, save for the most direct manner, to communicate to the guy she loves him, but the guy "just doesn't get why she's acting that way" and doesn't bother to question the behaviour any further.
Unrelated to your comment, but I really don't see the chemistry between Mishima and Yoshida. Almost every conversation we see between the two of them is either her criticizing his behaviour (which to be fair, he does have low self-esteem and is patronising) or him patronising her ("Re-do it." Like, I get that this is his job as he's training her, but how is a person ordering you around remotely charming?)
