Most likely the curse. At the beginning, they most likely ingested the flesh of the first priestess to start the curse.
Tradition to keep a baby’s umbilical cord as a good luck charm for the baby also had existed for centuries in Japan. So at the time of the first priestess, her family would have preserved her cord for something unrelated to the curse.

In case someone asked how could they prepare and store the umbilical cord needed for the ritual even before they created the curse.

Simple way to see it, after they found the second generation didn’t inherit the power, they start looking for way to artificially create the priestess, and happen to remember they had something from the original stashed away, and that can be used as catalyst.

So, uh, how many umbilical cords do you suppose Mirai had to eat to make a curse that strong?

The woman must've seen that line appear on the pregnancy test and just immediately chowed down on the world's largest bowl of uterine menudo.
i am more curious how many souls her child has eaten to get that strong. I have a feeling thats what happened to the moms soul.

Ah, for those wondering, its possible the umbilical cord of the first priestess was still around. It's rare, but some families have a custom of keeping the umbilical cord from children's births, for various reasons.

Might be a dumb question but couldnt they just kill her? The curse inside her shouldnt be able to survive without its “host”

An old woman died from her soul being ripped out of her body because Nana got near to her.
People in the know will not approach and people not in the know will report to the police of someone planning a murder.

If other popular supernatural battle manga is to be believed, curse tend to become more powerful with more limitation. If Mirai’s curse is specifically blended to “kill all Ebisumori (and only them)”, it might not immediately trigger against other people getting close.

My theory is that she ate her own(after Kaya) which started some corrupted form of the ritual or an infinite loop of curses
