He's been dodging any inquiry regarding whether he harbors feelings for Ayano, but this statement somehow convinces him to do the thing that might (it won't, but he's overly self-conscious and would think it would) implicate him?

From my interpretation, his sister is needling him about the sense of distance that he's trying to establish with Ayano. Like, 'of course you wouldn't know her personal plans, Isshiki-san' (with the san-suffix makes it come off as formal)
