So he wasn't reacting to Batman so much as he was reacting to getting caught up in Morino's flow.

A small part of the gag is Mejiro's Kansai-ben, "That's not a vampire, that's Batman" line (やなくて / やないかい is a distinctly Kansai negation/tag-question form), but we didn't think it was necessary for the joke to land, so we didn't try to add an 'accent' of some kind to the translation.

Mejiro spots the vampire-ish drawing in his notebook and reflexively blurts out a joke - "That's not a vampire, that's Batman" - which is a tsukkomi, the straight-man callback/correction move his dialect and backstory already told us he swore off doing with Tokyo people after bombing on his first day. He didn't mean to make the joke. It just slipped out, naturally, unprompted - specifically with Morino. That's what "I've done it now-!!!" is actually about on the very next page: he's not panicking over Batman; he's panicking because he caught himself falling into an easy, reflexive comedic rhythm with her that he told himself he'd never let happen again. The Batman line is the evidence of the slip, not the joke itself.

So the beat is: his defenses came down around Morino without him noticing, and the tsukkomi is the proof. It's an "oh, that's sweet" reveal rather than "he's embarrassed about a doodle".
