Are you being serious? Comedy has existed in society for as long as society itself. We’ve seen graffiti in Pompeii ruins that attests to that.
Maybe access to it, though even that is doubtful. People would've still made jokes in pubs or front rooms before performing it on a bigger stage. If he means comedy club like performances then maybe I could stretch my disbelief. I doubt given the nature of this world there's been a big performance before.

Is it me or the nun lady kept getting bigger each chapter?
this is her first (somewhat) full body comparison with Mikagami in the bottom panel from chapter 21 (click to zoom)

In this chapter 24, you can see she is roughly the same size difference compared to Mikagami. In the panel when she protected the orphans, you can see she's not too big. Her proportions are the same in serious situations:

However, on panels featuring herself, the author follows the rules of comedic exaggeration, making her look larger in all aspects:

I don't think that's comedy at a theater level. Probably an inside joke in a community size of comedy
And definitely not the level of Japanese slapstick comedy. Those can get ridiculous high and fast

Dirty joke is a culture that are shared across time and places
Comedy didn't probably develop especially Dirty Jokes, might be related to the fact that the Colosseum is in a Religious Nation, who had been in constant war with neighboring nations. When everyone is focused on just war, people tend to ignore other stuff, other than fighting and physical desires.
