Yotsuyu was an actual spider who lived long enough that one day she'd just found herself turned into a human woman's form (and apparently magically knew how to speak Japanese). I'd be surprised if she ever met her parents, and it's unlikely they also were 'promoted' to jorogumo. But who knows - whatever works for the plot! I think you'd be right in the yokai case.I wonder if Yotsuyu's parents really care or if she can even find them. Maybe being humanoid, mythological spiders makes things different, but I still reckon there might not usually (naturally) be actual couples. The pair might just have a short affair, the male would go away (could be chased away by the female) and the female would then take care of the child/children alone, just long enough for them to survive on their own. Then the children would go their own ways, never to come back. That would sound about right if you asked me. However, being taught by Toda could make Yotsuyu change enough to accept Toda hanging around, even if they became a couple.

That's the interesting thing about Yotsuyu to me. There's no indication she comes from a family of monster spiders. I assume, just as Charlotte's Web tells us, her father and mother died before or shortly after she sailed away from the egg sac on a silken thread. The specific magic of being a yokai seems to have prevented her from being completely feral, but she's not human. She's a completely singular existence now integrating into human and yokai society.
