I think it is because they bite him while he is weaken. While there is a connection that MC can feel their emotion, there must also be some link they can do something to MC as well.

I am excited to see where the story goes given the author of the game is now working on the manga aswell.

If you're interested, in addition to the main story, there are now also spin-offs with alternative storylines:

First is "Tokiko IF STORY" spin-off manga (drawn by different artist), which the original author writes in conjunction with mainline storyline (both mangas are now published bi-monthly, so one month for main story and the next month for "Tokiko IF STORY" and so on). “Tokiko IF STORY” is based on a simple premise "What if Tokiko hadn't turned into a zombie?" and offers complete alternate storyline following that premise. Unfortunately, it hasn't been translated yet, but considering that KDT SCANS has now caught up with the manga with this chapter, I wonder if they now could move their focus on "Tokiko IF STORY" next.

Second is "Road to Heaven" RTS x Tower defence game, which is currently in the works by TENDA Games. Nothing detailed known about it's story yet, but the description says "In this alternate timeline, characters who never met in the original story, former enemies, and even those who had already died may become your allies. Will you reach out and help others? Will you make ruthless choices to keep your own group alive? The kind of survivor Yusuke Takemura becomes is up to you." The game has a confirmed official English release and you can already add it to your wishlist on Steam.

It's not really route from VN adaptation. It's a completely alternate scenario straight from beginning with human Tokiko as heroine and diverging completely from original story because of that.

I don't really see how such an alternative scenario could really be made for either Tsugumi or even Sayaka, considering how much later they appear in main story. But who knows.

Sayaka just dont seem popular enough to ever star. Even in vn vol.2(which is kinda her arc), she never got any form of "romance"(and i use this liberally, relative to vol 1)

For all of Yusuke's guilt( his failure to protect Mitsuki and her brothers as well as his (stated in game) towards Sayaka for knowing that she wants a serious relationship despite saying she is fine with a physical relationship only with him indulging in it), his relationship with the 2 ladies do end up very different.

vol 2 and 3 each have the option of a tokiko ending, and iirc, a fantl(of the web novels) translation of author word did mention author liking tokiko personally.
But yeah, Mitsuki is the heroine of the main timeline.
None of them ever have a Sayaka ending iirc.

Well, with Tsumugi pretty much disappearing(even though we got teased with a h extraa chapter), I doubt she would ever get popular enough to star(and the author probably use her as a "bait/shock" factor i suppose. idk why but she does remind me of Mitsuki so I was like, this being a h series would feature her right? yet how would the author excuse MC from cheating on Mitsuki blah blah blah) tl;dr rip Tsumugi. Really wished for her.
I suppose, in the same vein, current arc is meant to establish Mitsuki as "THE" main heroine, by having tokiko being evil so to speak(she and other zombie girl maybe maybe not be forming a separate zombie faction from the main antagonist, but that doesnt excuse their pretty much massacre of a human group). So Yusuke might eventually have to put tokiko down.
