Let's look at Akane and her feelings for Sou. She has an iron will that prevents her from going overboard but Akane herself is already an outlier, both as a woman and as a mother. Why? Because of Sakura - just her being there is unusual. Most boys never have a younger sister since women usually stop having kids once a son is born. So males never take care of younger siblings, instead it's always them getting cared for, confined and spoiled rotten - probably even lusted after.
That also explains, as mentioned, why Sakura would seem different from other girls her age. If you are born a girl, then you live in a family with your mother and probably have a sister or two. The usual girl doesn't have a brother, let alone a father who lives with her. Obviously contact to males is limited or even zero. Girls having fantasies is only natural. But that's where Sakura is not normal. First of all, her family has a boy - that in itself is unusual, but she's an even greater outlier since Sakura is a girl who never experienced life without a male near her. That in itself sets her miles apart.
Sakura being the "little sister"of a boy would obviously have less illusions about guys than your average girl from next door. That being said, it means having a normal boy (old Sou) disillusioned her about guys almost from the moment she was born. And here comes current Sou, treating Sakura sweetly, being a chad. cooking for his little sister, patting her and caring for her like the imaginary Onii-chan all girls in her class dream of at night. How blessed can she be? Of course Misaki would notice a change in her friend's behavior.
