Never specified, I think? First chapter was Nana executing her plan already. As far as I can remembered, she just recognized he was a good candidate and she wanted him. It seemed to imply she started catching a feeling for him properly when she visited his house (when Mana was conceived), got to see his face properly, and realized Ryuuji was actually hot.
