That flower's meaning was "I bid farewell to the world without you", and we saw it from Ameri's mother's background in chapter 327 too.(Also that volume's inside cover) So I think that door leads to human world.

Maybe Ameri might use that door when Narnia and Baal banishes Iruma back to human world, as she bids farewell to the world without Iruma.

You're getting too worked up here. Sure, maybe the chapter end is a red herring and they won't actually go to the human or any other world. But there's a non-zero chance they will.
You're arguing on the assumption that the gate should be working if it was real, which would go against established world-building, so it can't be real.
But you're not thinking of the possibility that the gate might just be no longer functioning, so no one knows where it was supposed to lead, if it ever worked at all. Iruma getting into contact with that gate could be the trigger to make it work again. Could be because he's human, could be because his ring is filled with Sullivan's mana—powerful enough to operate the gate—or it could be because his ring is inhabited by Ali who is so far assumed to be the previous Demon King Derkila. Wherever that gate supposedly leads, there's a good chance Iruma is the only one who can open it.

Saying that the door couldn’t possibly have any connection to earth is like saying Knowledge can’t be lost over time. at One point the door might’ve led to the human world, but after eons of it being sealed shut, all that’s left is an old myth that it goes to another world. if Iruma is gonna end up opening it this arc, maybe it required a human to activate it and that’s why nobody remembers/knows where it goes

I guarantee you the portal works, but it requires human activation or something, and when Iruma touches it he accidentally takes Asmodeus and Clara to the human world, queue shenanigans, he tells them he's human, they love him anyway, they go back, and it draws a bunch more eyes on iruma and it leads into his final reveal and ultimate victory over the netherworld.
