Not necessarily true. Shoujo manga/anime with a male lead do exist. Aishiteruze Baby is an example.

It was listed as a shonen, but the series got transferred to Hana to Yume+ (The new website for the shoujo magazine, Hana to Yume that includes other original series) a few weeks ago, and even Hana to Yume included the series in their announcement tweet, and it was part of the Hana to Yume 52th anniversary celebration.

It's a similar situation to series like JoJo's part 7 or Vinland Saga (both started as shonen but then got transferred to a seinen magazine), so now it would be a shoujo.
