Considering this is a relatively wholesome manga, I think it's the second prince who was friends with Sophie. Once they're married to their respective princes, they'll become real sisters or something wholesome like that. Second prince has no interest in the throne so it makes sense he can goof around with Sophie in their childhood as well.

I don't think Onee-sama have that much ill-will towards Sophie, she's genuinely interested in her and I presume making Sophie her little sister might be her way to gauge Sophie's character (a test, if you will). She just said the "aren't you mad at me" thing because she feels guilty afterward probably.
