The Three Heroes are basically those who had been experimented on and infused with Diablos blood/genes, and a sign of that Diablos blood is the condition of "Possession".
So far, girls with Possession have either been killed-off or taken into custody by the Church or the Cult (were they are either experimened on or discarded).
Leaving those aside and the ones related to the Royal blood around the world or whose blood haven't reacted yet to Possession, the majority of Heroes Descendents/Diablos blood carrier are in Shadow Garden (around 700 of them).

exactly why i said once is coincidence and twice is foreshadowing. first hero having the same facial features of alpha is a coincidence but the cat hero essence having the same as zeta is beyond coincidence regardless if they're the possessed. like what are the chances that the seven shadows have also the same facial features of the heroes of forgotten history?

Akane is from Earth, so she has no relation with Diablos or the Three Heroes.

And I am not sure whom you are referring as Pink-haired Yandere under Zeta, because I only know of one Red-haired Human girl (as per LN art) who works under her, and she doesn't seem like a Yandere, atleast what's shown of her in the Manga.

The only Humans we know who have shown signs of Possession are Claire, Nu, Rose & Millia.
Alexia haven't shown the signs, but the Cult had confirmed that she along with her sister Iris belongs to the Human Hero bloodline.

Pink hair is presumably referring to 559. Probably isn't aware she's an elf, not a human.

But question is who directly represents those Heroes.
Alpha represents the Elf Hero.
Zeta seems to represent the Beastman Hero.
That leaves the Human Hero as the last one, but Shadow Garden doesn't have that many Human members and none of the Seven Shadows are Human.

Both Gamma & Epsilon are Elves.
The only known Humans in Shadow Garden are Nu & Rose (as per Manga).
The rest known ones are either Beastgirls or Elves or Dark Elves.

I think the reason there are so few Possessesed affected Human girls in Shadow Garden is because the Cult and the Church has a higher influence on the Human society, unlike the Elves and Beastman society.
While all society heavily discriminates girls affected by Possession, the other society either just abandons them or kill them or sell them, while the Human society just hands them to either the Church or Cult, were they either get experimented on or slaughtered.
