She's not autistic, but her ability/inability to deal with kids isn't why. My wife is diagnosed with Level 1 Autism (high-functioning, but may need mild support in social situations) and she's amazing with kids. Autism has issues recognizing non-verbal cues when it comes to learned social behaviors, my wife described it as "it's like everyone else got a rulebook except me."
Children often haven't learned these non-verbal social behaviors so their non-verbal cues are pretty honest and blunt with what they're feeling, something someone with autism may resonate with, or they may not as it really depends on the individual.

But yeah, Koyuki doesn't have autism.
