Eh, I've seen bigger be done perfectly fine, it's just that the proportions here are all off. The shoulders are way too broad, and Neiro's jawline has completely changed shape for no reason. Obviously, the author made Neiro look extra mannish because that's the joke, but if the author were interested in doing "buff Neiro" as more than just a one-page gag, I'd suggest something more like this piece by SheepApp:

This girl's arms are actually about the same size as page 14 Neiro's relative to the size of their heads (obviously we're speaking proportionally here), but the shoulders are kept a lot more narrow, keeping the body shape a lot more feminine despite the buffness. It's a good example of how an artist could put a lot of muscle on a body type like Neiro's without making her look like a totally different person.

Yup. Skeletal structure matters a lot and in most cases, much more than even one's muscular structure. Men and women actually have very, very similar musculature, so fat deposits aside, most of our sexual dimorphism comes from mr./ms. skeltal.

A woman with the musculature of an average (in decent shape) male would mostly just look like a slightly yoked woman with a more built upper body (and possibly strange hips). That's why just changing the template anywhere tends to look so instinctually strange.
